The most important thing for me is "to thy own self be true".
What that statement means to me is to know what my values are. Have a moral standard that I live by the best I can by not just talking about it but I try to walk it. Moral character is another. I sometimes fail at my walk but I keep trying to achieve it.

I too believe it's important to stick to your values and moral standards/character but I also believe we have to forgive ourselves when we fail, no one is perfect.
With that in mind, what are the most important things to learn about one’s self?
For me, it was learning what was wrong with me - receiving a diagnosis of my mental health - and understanding why I had these mental problems. Then I was able to proceed from there in my recovery. I had been spinning my wheels for decades while trying to deal with my problems without understanding what they were.

First is that whoever you are is good enough. Second is that it is not set in cement. We can change and grow.

I hope that struggle becomes much easier with time. To be honest, I'm not sure if we ever will know ourselves completely, but we can come close by "showing up for ourselves", to listen what's going on inside. Mindfulness can help with it.
Use one's strengths, work on your weaknesses and introspect every once in a while.

Look at myself carefully and see when I am telling myself things that are not true. That tends to happen when I create false hopes of happiness with things that lead to suffering

Good point soulsings. What I tell myself is what my mind believes so I need to be mindful with how I see things and with choosing my words.

To truly understand a person we need to learn about them by listening mindfully in my view.
